Part 1 of 4Requirements for Growth
To test substances that kill microbes or stop their growth, the bacteria must first be given everything they need.
Nutrients
- Nutrients Supply the materials needed for growth and a source of energy for respiration.
Oxygen
- Oxygen A sufficient supply is needed for bacteria that respire aerobically.
Temperature
- Temperature It must not be too high or too low, so that enzymes controlling metabolism work close to their optimum.
pH
- pH An unsuitable pH changes the shape of enzymes and slows or stops the metabolic reactions.
Water
- Water Moisture is needed because metabolic reactions take place in solution inside the cells.
Part 2 of 4Providing the Conditions
Providing the Conditions
Broth
- Broth A liquid of distilled water and nutrients in which a bacterial culture can be grown before use.
Agar plate
- Agar plate A petri dish of agar jelly acts as a solid growth medium holding the nutrients.
Incubation
- Incubation Plates are incubated at a suitable constant temperature for 24 to 48 hours to allow visible growth.
Why 25 °C
- Why 25 °C In schools a temperature of about 25 °C encourages the bacteria without promoting the growth of human pathogens.
Part 3 of 4Why Control Matters
A fair test of an antimicrobial substance needs identical conditions for every sample.
1
All agar plates should be inoculated with the same culture in the same way and at the same density.
2
Temperature, pH and nutrient supply should be the same across all plates so they do not influence growth.
3
Any difference in growth can then be attributed to the substance being tested.
Part 4 of 4Exam Checklist & Tips
4.3.1 – Key Points for the Exam
- Broth A liquid medium containing water and nutrients for growing microorganisms.
- Agar A jelly used as a solid growth medium in a petri dish.
- Incubation Keeping cultures at a set temperature so that they can grow.
- Aerobic respiration Respiration that needs oxygen.
Common Mistakes
- Incubating at body temperature in school, which may encourage growth of harmful human pathogens.
- Forgetting that enzymes control bacterial metabolism, which is why temperature and pH matter.
- Thinking all bacteria need oxygen, when this applies to those that respire aerobically.
- Allowing different plates to have different conditions, so results cannot be compared fairly.
